I just saw the original The In-Laws after a long time and feel that Peter Falk and Alan Arkin form the best buddy-buddy pair ever.

Robert De Niro and Charles Grodin in Midnight Run are a very close second.

I did not like the Eddie Murphy & Nick Nolte pairing in 48 Hours. I dislike both actors (especially Murphy) and the film was nothing special.

For the "buddettes" Susan Sarandon and Geena Davis were excellent in Thelma & Louise

Just by having 2 same sex actors as friends or antagonists does not necessarily make them a buddy pair. Some Like It Hot is one of my favourite films and both Jack Lemmon & Tony Curtis were excellent but being in the same plight together, they do not make perfect foils for each other - something that is essential for real buddy-buddy films.

Kilmer and Downey jr in Kiss Kiss Bang Bang gave me some hope there would be a revival in buddy/buddy movies.

John Belushi & Dan Aykroyd were a good pair in The Blues Brothers but the cameos by the supporting cast and the film itself took a significant share of the honours, thus diluting the 'buddy effect'.

I came in here to post Gunga Din (1939)...and saw I'd posted exactly the same 5 and half years ago...

Not seen it for a while but as a lads movie it made me laugh a lot.
